Rats with chronic and progressive overfeeding are used as a model for the study of several aspects of obesity and the development of insulin resistance. Overfeeding has been shown to decrease the pancreatic glucagon release and in turn to increase the insulin release. Although the overfeeding is supposed to improve the hepatic metabolic status, the direct effect of the overfeeding on the hepatic lipid metabolism has not been studied. In the present study, we used surgical Roux-en-Y gastric bypass (RYGB) and sham-operated rats to evaluate the effects of chronic overfeeding on the basal and glucose-stimulated rates of hepatic and pancreatic triglyceride (TG) secretion. The intraportal infusion of glucose (0.5 g/kg) in anesthetized animals was performed, with (RYGB) or without (sham) the gastric bypass. In RYGB rats, basal and glucose-stimulated hepatic and pancreatic TG secretion rates were lower than those in sham rats. In the sham rats, the basal and glucose-stimulated hepatic TG secretion rates were higher than those in RYGB rats. In the sham rats, the chronic overfeeding did not affect the basal and glucose-stimulated hepatic TG secretion rates. In the RYGB rats, the basal and glucose-stimulated pancreatic TG secretion rates were lower than those in sham rats, but this was only significant in the basal state. In the sham rats, the chronic overfeeding increased the basal and glucose-stimulated pancreatic TG secretion rates. In the RYGB rats, the chronic overfeeding did not affect the basal and glucose-stimulated pancreatic TG secretion rates. Our data indicate that both the basal and glucose-stimulated hepatic TG secretion rates are lower in RYGB rats than in sham rats.
